According to the error message :
it looks like the folder is not found.
Thus, it might be unrelated to pmwiki.
if I were you, I would do easy test :
just upload a basic html file
(http://fr.wikipedia.org/wiki/Hello_world#HTML) and call him
check if there is htaccess elsewhere than in pmwiki
how long have you been owning this domain/host ?
